No. You need to set the transparency so that the gray checkered background is visible (select it using the lasso/magic wand, press Del). By the way, converting is not necessary; Paint.net should open DDS files automatically.

Yes, if you put the tga file in the Complete folder, create a new Texture object in the editor, specify the name of this file in src, click on the new texture in the directory tree, right-click, Hierarchical->ForceExport, a new dds texture will be created. But the image size must be a power of two.
