Basically, the problem is this...

I can't use runes in the 2nd mission of the 2nd campaign... In the spellbook, they are gray (not highlighted), and when my squad's turn comes, the list of runes appears slightly above the chat... but when I click on them, nothing happens. I naturally have the resources, and I also take into account the requirements for specific runes.

Please help me figure it out.

This is an old problem related to the unwillingness to read the manuals ;) The solution is simple - during the turn of the dwarven unit, pay attention to the Action Points (AP) bar - the runes that the unit can activate will be displayed above it (and the player decides whether this activation is needed).
