If Kane was infected on the first night, he will die on the second night, unless he was infected twice that night, cured, and then recruited. Too many coincidences, in my opinion. Jen Li definitely wasn't infected, because on the first night, nothing could have removed her protection before Lucy was infected. I'm leaning towards the fact that the Maniac was infected. And his ultimate ability used on Rick on the second night with the infection is like an impudent gesture towards the town, which deprived him of his chances of winning on the first night. The Maniac could have been stunned by Rick, or blocked. Or, if we assume that he is infected, he had already used up both of his attacks by that point. But then, in theory, he should have used his recharged date ability, which he didn't. So, either the Maniac gave up on the game, realizing that he was doomed, and didn't send his special ability, or he was blocked/stunned.
I think that the infected Yaschur used his ultimate first, so with his last action, he stunned the Kosha-maniac.
