I think that if you approach everything with a lighthearted and humorous attitude, there won't be any conflict. Although, a lot depends on the players themselves.

Is the number of players fixed?
No. You can have ten players or a hundred. The main thing is that it's interesting and reasonably balanced.
What is the ratio between the groups of players?
One Mafia team should be approximately two to two and a half times smaller than the entire town. The number of Mafia teams is from one to three. If the setting is small, it's better to have one; if there are many players, you can introduce two Mafia teams. The number of serial killers and town roles is up to you. But if the setting is small and there are few players, it's better not to include town roles. And you also need to monitor the balance! This concept is, of course, quite mythical, but you should at least roughly ensure that the teams have something to counter each other and can act more or less on an equal footing.
